The Baltimore area has been inhabited by Native Americans since at least the 10th millennium BC, when Paleo-Indians first settled in the region. A Paleo-Indian archaeological site and several Early and Woodland archaeological sites have been identified in Baltimore, including four from the Late Woodland period.

In the early 17th century, the area around Baltimore was populated by Native Americans. Baltimore County in the north was used as a hunting ground by the Susquehannocks who lived in the valley of the lower Susquehanna River who "controlled all the tributaries of the Chesapeake" but "refused much contact with the Powhatan of the Potomac."
Oppressed by the Susquehannocks, the Piscataway tribe of Algonquians were south of Baltimore County and their main residence was the north bank of the Potomac River in what is now Charles and Prince George south of the Fall Line.
As shown in John Smith's 1608 map that faithfully mapped the settlement, there was no map near Baltimore, while noting the settlement of the Patuxt River below the level of the Piscataways.
In 1608, Captain John Smith traveled 210 miles from Jamestown to the upper Chesapeake Bay, leading the first European expedition to the Patapsco River, a term used by the Algonquin natives who fished and hunted.

The name "Patapsco" comes from pota-psk-ut, which translates to "backwater" or "tide covered with foam" in the Algonquian language.
A quarter of a year after John Smith's voyage, Lord Baltimore led 140 colonists aboard the merchant ship The Ark to settle in North America. The colonists were initially intimidated by the Piscataway in southern Maryland because of their body color and war toys, mistakenly assuming they wanted to attack the fleeing colonist's settlement. But the chiefs of the Piscataway tribe were quick to grant the colony permission to settle in Piscataway territory, and permanent relations were established between them and the Piscataway.

The County of Baltimore was "constituted" about 1659 in the records of the Maryland General Assembly as one of the first divisions of the Colony of Maryland into counties ordered to be served by the "Sheriff of Baltimore County." The area that makes up the modern city of Baltimore and its urban area was settled by David Jones in 1661, his claim included an area known as East Harbor on the east bank of the Jones Falls River, which flows south into Baltimore's Inner Harbor. The following year, shipowner Charles Gorsuch settled at Whetstone Point, Fort McHry's last site. Another "Baltimore" existed on the Bush River as early as 1674. The first county seat of Baltimore County is known today as "Old Baltimore". It was located on the Bush River on land that in 1773 became part of Harford County. When Baltimore on the Patapsco River was founded in 1729, Old Baltimore was gone.
The General Colonial Council of Maryland created and authorized the port of Baltimore in 1706 at the head of the northwest branch of the Patapsco River in what was later called "the Basin" (today's Inner Harbor) and later expanded east and southeast down the river. The settlement later known as Fells Point went east near the mouth of Jones Falls and continued in the nineteenth century to what became known as Canton.
Cecilius Calvert was the eldest son of Sir George Calvert (1579–1632), who became the first Lord Baltimore of County Longford, Ireland in 1625. The Maryland General Assembly established Baltimore City in 1729. Unlike other cities established during that time, Baltimore City was more than just on paper. German immigrants began settling along the Chesapeake Bay in 1723 and settled in Baltimore County. The General Assembly expanded the city of Baltimore in 1745 and incorporated the original settlement of David Jones called Jones Town. Baltimore's st representative of the congregation, and over the next two decades it acquired nine parcels of land and nearby villages that have been retained including Fells Point to become an important community at the head of the Patapsco River. As the town grew, an increasing number of German Lutheran immigrants founded Zion Church in 1755, and later the German Reformed congregation was organized for the first time among Protestant Protestants, which also attracted more of these "Pennsylvania Dutch" settlers to the region. Early German settlers also founded the German Society of Maryland in 1783 to promote the German language and German culture in Baltimore.
